A proof of concept methodology to identify protein structures that could be converted into engineered protein scaffolds.
We present a proof of concept methodology to identify protein structures that could be converted into engineered protein scaffolds, stable proteins whose sequence can however be mutated. These protein scaffolds are especially attractive for real-life applications, e.g., regulation of signaling pathways in vivo, therapeutics, and molecular imaging. By methods of directed evolution, we experimentally proved that the newly designed protein scaffold could be adapted into a nanomolar-affinity binder of human interleukin-10, a medically important target.
